The Human Immunodeficiency Virus Type-l Tat Protein Upregulates Bcl-2 Gene Expression in Jurkat T-cell Lines and Primary Peripheral Blood Mononuclear Cells

چکیده
The regulatory Tat protein of human immunodeficiency virus type-l (HIV-1) exerts a pleyotropic activity on the survival and proliferation of different cell types in culture. In this report, we investigated the effect of either endogenous or exogenous Tat on Bcl-2 proto-oncogene expression and cell survival in Jurkat T-cell lines and primary peripheral blood mononuclear cells. Stable and transient transfections of Jurkat cells with the cDNA of tat and a plasmid containing Bcl2 promoter in front of CAT (Bcl-2 PrlCAT) stimulated CAT activity and showed an increase of Bcl-2 mRNA and protein expression. This effect was specifically related to tat, because Jurkat cells transfected with the cDNA of tat in antisense orientation, tat carrying a mutation in the amino acid cysp-glp, or the control vector alone (pRPneo-SW) did not show any significant difference in Bcl-2 promoter activity with respect to parental Jurkat cells. We also observed a specific correlation between tat-induced Bcl-2 gene expression and inhibition of apoptosis induced by serum with-

متن کاملUpregulation of c-Fos in activated T lymphoid and monocytic cells by human immunodeficiency virus-1 Tat protein.
The regulatory Tat protein of the human immunodeficiency virus type-1 (HIV-1) is essential for viral replication and also shows pleiotropic activities on various cell functions. To get further insights into the molecular mechanisms underlying the biological activity of Tat, we investigated the effect of endogenous and exogenous Tat protein on c-fos gene expression in T lymphoblastoid (Jurkat) a...

Introduction: Interleukin-2 (IL-2) as a T helper type 1 (Th1) cytokine has an important role Background and Objective: Interleukin-2 (IL-2) as a T helper type 1 (Th1) cytokine has an important role in activation, growth and differentiation of several immune cells. Moreover IL-2 is known as a pro-inflammatory and anti-tumoral cytokine. In addition deysregulation of IL-2 in some diseases such as ...
متن کاملTat protein induces human immunodeficiency virus type 1 (HIV-1) coreceptors and promotes infection with both macrophage-tropic and T-lymphotropic HIV-1 strains.
Chemokine receptors CCR5 and CXCR4 are the primary fusion coreceptors utilized for CD4-mediated entry by macrophage (M)- and T-cell line (T)-tropic human immunodeficiency virus type 1 (HIV-1) strains, respectively. Here we demonstrate that HIV-1 Tat protein, a potent viral transactivator shown to be released as a soluble protein by infected cells, differentially induced CXCR4 and CCR5 expressio...
متن کاملExpression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide
Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...
